r-HuEPO tedavisi ve serum total glutatyon s-transferaz aktivitesi

Abstract

Glutatyon S-transferaz (GST)'lar organizmada yaygın olarak bulunan bir enzim grubudur. Bazı hastalıkların ve hormona! manipıılasyonların bu enzimin hücresel düzeyini değiştirdiği bilinmektedir. Hu çalışmada diyaliz hastalarında r-IIuFPO tedavisinin serum total GST konsantrasyonuna etkisini inceledik. Hastalar üç gruba ayrıldı. Grııp-I en az altı aydır haftada 2000-4000 II ’ sııbkutan r-Huh.PO tedavisi gören 15 hastadan oluşuyordu. Grup-II kontrol grubu olup hiç r-lluFPO tedavisi almamış 15 hasatçıdan oluşuyordu. Grup-III'de ise çalışmadan önce r-UuEPÜ tedavisi görmemiş 10 hasta mevcud olııp bu gruptaki hastalar 3 aylık çalışma süresince haftada 3000 11'subkuian r-HuFPO tedavisi aldılar. Gruft-Iir deki hastalardan tedavi öncesi ve sonrası serum GST aklivilesine bakıldı. Grııp-I ve -II’ deki hastalarda sadece bırkez GST aktivitesi bakıldı. Hastalarda total GST tayini llabig ve ark.' nın metoduna göre çalışıldı. (irıtp-I ve -//' deki hastalarda serum GST aktivitesi açısından anlamlı bir fark yoktu (p 0.1). Fakat Grup-III' deki hastalarda r-lluFPO tedavisi öncesi serum GST aktivitesi üç aylık r-IIl’FPO tedavisi ile 31.69 ± 6.23 I I.' den 39.04 ± ~.20 l' L düzeyine yükseldi Bu istatistiki olarak anlamlı idi (p 0.03). Ayrıca serum GST konsantrasyonu ite serıım transaminaz düzeyleri arasında anlamlı bir korelasyon ve HBs hastalarla HBs - hastalar arasında anlamlı bir fark tespit edilmedi. Sonuç olarak. r-IIuFPO, tedavinin başlangıcında serum GST seviyesini yükseltmekte fakat uzun süreli tedavi ile bu etkiye karşı tolerans gelişmekle olduğu sonucuna varılabilir.

GST activities were ana/yzed in the beginning of the sludy. ıı hereas in Group-1 II these assays ere performed both at the onset and in the end of three months' sludy. GST assays ere done by the method of llabig el al. There vas no signifıcant differances between groııp-I and -II in lerms of serum total GST activities (j> 0.1) but GST activities in group-III w ere increased with r-lluFPO treatment in the end of sludy (39.04 ± ~.2 1' 1.) compared to the activities obtained at the beginning of the treatment (31.69 ± 6.6) (/> 0.03). On the other hand we could not Jind anv correlation beiween serum GST and Transaminase activities and any dıfferance between GST activities of HBs • and HBs-patients. In conlusion, r-lluFPO mav increase the level of serum GST activity with short-term treatment (3 months). but m ay gain tolerans with loııg-term r-lluFPO treatment.
