Kırmızı şarapta 14 fenolik bileşiğin eş zamanlı tayini için ters faz yüksek performanslı sıvı kromatografisi metodu geliştirilmiştir. Belirlenen bileşikler, gallik asit, kateşin, 3,4-dihidroksibenzoik asit, klorogenik asit, epikateşin, 4-hidroksibenzoik asit, şiringik asit, kafeik asit, p-kumarik asit, rutin, resveratrol, mirisetin, kuersetin ve kampferoldür. Sıvı-sıvı ekstraksiyonu metodu, katı faz ekstraksiyonu metoduyla karşılaştırılmıştır. Metot asidik pH'da etil asetatla sıvı-sıvı ekstraksiyonunu içerir. Zorbax Eclipse XDB-C18 kolon (5µm, 4.6mm x 250 mm) kullanılmıştır. Bileşiklerin kromatografik ayrımı %0.2 formik asit içeren metanol su karışımıyla çözücü programlaması yapılarak, oda sıcaklığında, 1 mL dak-1 akış hızında gerçekleştirilmiştir. Tayin hem UV-GB dedektörüyle dört farklı dalgaboyunda, hem de floresans dedektörle gerçekleştirilmiştir. Her analizin tamamlanması için 14 dakika analiz süresi ve sonrasında 10 dakika dengelenme zamanı gereklidir. Optimize edilmiş kromatografik metot valide edilmiştir. Floresans dedektör kullanımı, kateşin, epikateşin ve resveratrol için artan seçicilik ve duyarlılık sağlamıştır. Sonuç olarak, tanımlanan bu metot, yöresel kırmızı şaraplarda fenolik bileşiklerin tayininde başarıyla uygulanmıştır.
A reversed-phase high performance chromatographic method for simultaneous determination of 14 phenolic compounds in red wines was developed in this study. The identified compounds contained gallic acid, catechin, 3,4-dihydroxybenzoic acid, chlorogenic acid, epicathechin, 4-hydroxybenzoic acid, syringic acid, caffeic acid, p-coumaric acid, rutin, resveratrol, myricetin, quercetin and kaempferol. The liquid-liquid extraction method was compared with solid-phase extraction method. The method includes liquid-liquid extraction of acidic pH with ethylacetate. The analysis used a Zorbax Eclipse XDB-C18 column (5µm, 4.6mm x 250 mm). The chromatographic separation of these compounds performed in a single run by using the mobile phase gradient elution of methanol water mixture ( % 0.2 formic acid) at room temperature, with flow rate at 1 mL/min. Detection was carried out by UV-Vis detector at four different wavelengths and fluorescence detector. Each analysis required an equilibration period of 10 min and a run time of 14 min for completion. The optimized chromatographic method was carefully validated for precision and accuracy. The use of a fluorescence detector allowed increasing selectivity and sensitivity for the determination of catechin, epicatechin and resveratrol. Consequently, the described method was successfully employed to study phenolic compounds in local red wines.
