VIU - ÖZET İnönü Üniversitesi Tıp Fakültesi Deneysel Araştırma Merkezinde orta derecede alkol entoksikasyonunun rat deri fleplerine etkisini araştırmak için yapılan kontrollü deneysel çalışmada, 12 hafta süreyle modifiye sıvı diyet, alkol+sıvı diyet ve katı diyetle beslenen ağırlıkları 190-275 g olan erkek wistar ratlarda akut dorsal McFarlene deri flepleri kaldırılarak, oluşan nekroz ve kontraksiyon miktarları incelenmiştir. Hayvanların alkolik oluş derecesi kan alkol düzeyleri, davranışsal ve EYS parametreleri ile değerlendirilmiştir. Hayvanlarda gözlenen ortalama 195 mg/dl'lik etanol kan seviyeleri ve ciddi etanol yoksunluk belirtileri sonucunda orta derecede etanol entoksikasyonu oluştuğu kabul edilmiştir. Flepler kaldırıldırılıp yerine sütüre edildikten sonra 10. günde fleplerde gelişen nekroz ve kontraksiyon miktarları değerlendirilmiş ve hayvanlar sakrifıye edilmeden önce plazma SOD, katalaz, MDA ve eritrosit içi SOD, katalaz, MDA ölçümleri için kan örnekleri alınmıştır. Alkolik ratlarda eritrosit içi SOD ve MDA değerlerinin anlamlı oranda düşük olduğu gözlenmiştir (p <0,05). Plazma SOD aktiviteleri alkolik grupta daha yüksek olmasına rağmen, bu fark istatistiksel olarak anlamlı bulunmamıştır (p>0,05). Flep kontraksiyon miktarları süt diyeti grubunda diğer iki gruptan daha az bulunmuştur (p<0,05). Alkolik ratlardaki ağırlık kaybı diğer iki gruptan fazla gözlenmiştir (p<0,05). Bu sonuçlar alkolik ratlarda gözlenen deri flep yaşayabilirlik artışının antioksidan savunma sistem aktivasyonu ve / veya azalan portein seviyeleri ile ilgili olabileceğini, ve yağlı diyetle beslenmenin deri flep kontraksiyonunu arttırdığını düşündürmektedir. 41
SUMMARY In order to evaluate the effects of moderate level of ethanol intoxication on the acute Mc Farlene's skin flaps in rats, a controlled experimental study was conducted. 190-275 g weighed male wistar rats were used. At the beginning of the experiment rats were randomly fed with modified liquid, etanol containing modified liquid and standart solid diets. Acute dorsal McFarlene skin flaps were elevated and resutured to donor areas on the 84. th day. The criteria for ethanol dependence in rats was blood ethanol level, behavioral changes and ethanol withdrawal symptoms. The mean blood ethanol level of rats fed with ethanol containing modified liquid diet was 195 mg/dl. Severe EWS (ethanol withdrawal syndrome) score was determined as moderate level intoxication in the alcoholic group. After resuturing flaps to the donor sites, the amount of necrosis and contraction were measured and blood samples were taken to evaluate superoxide disfnutase (SOD), catalase and malondialdehyde (MDA) in plasma and erythrocytes on the postoperative tenth day. In the alcoholic group; SOD and MDA levels in erythrocytes were found in low levels (p<0,05). Despite the high level of plasma SOD activity in alcoholic group, it was not statistically significant ( p>0,05). The amount of flap contraction observed in modified liquid diet group was less than the other groups (p<0,05). Weight loss in the alcoholic group was statistically significant than the others (p<0,05). These results suggest that the increased skin flap survival observed in alcoholic rats may be due to protein depletion or/and activation of antioxidant defense systems. Feeding with high fat diet may increase skin flap contraction. 42
