Abstract:
Öz:T. vaginalis, insanın ürogenital sisteminde yaşayan, kamçılı bir protozoondur. T. vaginalis'in ürogenital sistemde parazitliğine Trichomoniosis denir. T. vaginalis fagositoz veya osmoz yolu ile lökositler, diğer vücut hücreleri, bakteriler ve vaginanın glikojeni ile beslenir. Hafif asit ortamda (PH 5,8), anaeorobik şartlarda ve 35- 37 ∘C de ürer. Boyuna ikiye bölünerek çoğalır. Trichomoniosis'in kesin tanısı ancak etkenin görülmesi ile konulmaktadır. Enfekte kadınların %10-50'si asemptomatiktir. Klinik bulgulara göre kadında veya erkekte, idrar ve üreme yollarının çeşitli hastalıkları ile karışabilir. Direk mikroskobik bakı ve kültür yöntemlerinin birlikte kullanılması duyarlılığı %98'lere kadar arttırmaktadır. T. vaginalis'te kullanılan kültür yöntemleri; CPLM (cycteine- peptone- liver- maltose) CPLM, Modifıye CPLM, VF Buyyonu, Diamond TPS-1 (Trypticase-pepton-serum) besiyerleridir. Çalışmada CPLM besiyerinde insan, at, koyun serumları kullanılarak, besiyerinde T. vaginalis'in üreme süreleri ve yoğunlukları karşılaştırılmıştır. İnönü Üniversitesi Tıp Fakültesi Turgut Özal Tıp Merkezi Kadın Doğum Polikliniğine gelen kadınlardan alınan vaginal akıntı örneklerinden izole edilen ve Parazitoloji Anabilim Dalında sürekli pasajlarla canlılığı sürdürülen T.vaginalis suşu kullanılmış olup çalışma sonucunda, insan serumu kullanıldığında üremenin at ve koyun serumuna göre daha fazla yoğunlukta ve yaşam süresinin ise daha uzun süreli olduğu gözlenmiştir. CPLM besiyerinde insan serumu kullanımının daha ekonomik ve uygun olacağı saptanmıştır.
Başlık (İngilizce):The influence of different sera on the reproduction and concentration of Trichomonas vaginalis in the CPLM media
Öz (İngilizce):T. vaginalis is a flagellated protozoon living in the urogenital system of human and is the cause of trichomoniasis. T. vaginalis feeds on leukocytes, other body cells and bacteria by phagocytosis and vaginal glycogen by osmosis. Under weak acidic (pH 5.8) and anaerobic conditions, they reproduce at a temperature of 35-37 ∘C. Reproduction occurs by lengthwise division. The exact diagnosis of trichomoniasis can be made only by seeing the organisms. From 10-50% of infected women are asymptomatic. The clinical picture in women and men may be confused with various diseases of the urogenital system. Direct microscopic examination and culturing may increase the sensitivity up to 98% if used together. The culture media used for T. vaginalis are CPLM, modified CPLM, VF broth, and Diamond TPS1 culture, hi this study, the length of the reproduction periods in CPLM medium containing human, horse or sheep sera were compared. T. vaginalis strains that were isolated from patients in the gynecology and obstetrics department of the Turgut Özal Medical Centre, Medical Faculty, Inonu University. These strains that had been kept alive by continuous passages by the parasitology department were used. Reproduction in human serum shows a higher rate and longer life compared to that in sheep or horse sera. It was found that the use of human serum in CPLM media is more economical and suitable.
